The Pixelgen Proxiome Kit offers the next omics dimension for single cells, protein interactomics
STOCKHOLM, Sweden and MARCO ISLAND, Fla., Feb. 24, 2025 -- Pixelgen Technologies today unveiled a novel technology for exploring the dynamic protein interactome at the single cell level, building on its earlier technological innovations in DNA-based 3D proteomics. The Proximity Network Assay delivers nanoscale spatial analysis of immune cell proteins at scale. The new technology underpins the Pixelgen Proxiome Kit, which launched today as the first kit in a new product line offering researchers the highest resolution of single cellular analyses by providing a new omics dimension for novel insights into disease mechanisms, drug response, biomarker discovery, and more.

At this stage of scientific discovery, new advancements will center around our understanding of proteins. Proteins do not act alone but as spatial constellations and interactions controlling the activities and functions of cells. The protein interactome represents the network of these functional interactions between proteins that drive essential biological processes, such as signal transduction, migration, metabolism, gene regulation, and immune responses. The Proximity Network Assay at the core of the Pixelgen Proxiome Kit allows researchers to study protein interactomics at an unprecedented scale.
The Pixelgen Proxiome Kit, Immuno 155 uses the Proximity Network Assay, a DNA-based chemistry where each target and its neighbors in close proximity are assigned a unique spatial position across single cells at an average resolution of 50 nanometers. Its validated protein panel contains 155 immune cell surface protein targets to produce a detailed map of up to 50,000 proteins per cell, illuminating cell surface protein abundance, clustering, and colocalization. The Pixelgen Proxiome Kit is instrument-free and easy-to-use, analogous to a sample-prep kit for NGS, and supports cells in suspension from PBMCs, BM, cell lines, and dissociated organoids.

The Proximity Network Assay uses DNA barcoded antibodies and rolling circle amplification, with the addition of proximity oligos and a gapfill-ligation reaction, to form multiple connections between neighboring proteins. The molecules are then sequenced to create a nanoscale protein Proximity Network for each cell, which is analyzed using spatial statistics to define the organization of each cell's protein interactome and cell-cell complexes.
